摘要
将48只SPF级雄性C57/BL6小鼠随机分为对照组、模型组、电针组、氟西汀组各12只,采用慢性不可预见性温和性应激制备抑郁样小鼠模型,电针组给予电针百会穴、印堂穴(连续波,2 Hz,1 mA,30 min/d),氟西汀组给予10 mg/kg氟西汀,每日干预1次,连续干预10 d。使用糖水偏好实验、强迫游泳实验、悬尾实验、三箱社交实验、Y迷宫实验评估各组小鼠抑郁行为、社交能力及空间记忆,使用离体电生理技术记录小鼠海马场电位,评估各组小鼠突触可塑性。
与对照组比较,模型组小鼠糖水消耗、社交时间、Y迷宫新奇臂进入次数与探索时间、fEPSP斜率均明显降低(P<0.01),强迫游泳与悬尾实验不动时间均显著增加(P<0.01)。与模型组比较,电针组、氟西汀组小鼠糖水消耗、社交时间和Y迷宫新奇臂进入次数及探索时间、fEPSP斜率均明显增加(P<0.05或P<0.01),强迫游泳与悬尾实验不动时间均显著减少(P<0.05或P<0.01)。与氟西汀组比较,电针组小鼠糖水消耗、悬尾实验不动时间、fEPSP斜率均明显降低(P<0.05),强迫游泳不动时间、社交时间和Y迷宫新奇臂进入次数及探索时间均无明显改变(P>0.05)。
抑郁症(depression)是一组以长期心境低落、愉快感缺乏、精力减退为主的常见精神障碍,全球每年约有5%的人患有抑郁症,而在中低收入国家更是高达80%~90%,且75岁时的抑郁症平均发病风险预计为19.6%,抑郁症已然成为世界范围内主要致残原因,也是全球疾病总负担的主要原
48只6~7 w龄SPF级雄性C57/BL6小鼠购于厦门大学实验动物中心,动物许可证号:SCXK(闽)2018-003。饲养环境:每笼4只小鼠,12 h/12 h光暗周期,温度(22±2) ℃。实验前将其适应性饲养1 w,随机分为对照组、模型组、电针组、氟西汀组各12只。所有操作均严格遵守厦门市仙岳医院实验动物管理伦理审查委员会的规定,符合动物实验行为规范(批准文号:2022-KY-013)。
盐酸氟西汀分散片(Patheon France,20 mg),异氟烷(麦克林,I833358-250 g)。
一次性无菌针灸针(中研太和,0.16 mm×7 mm)、实验动物麻醉呼吸机(瑞沃德,R58S)、刺激器(BMS-master-9)、小动物行为学检测系统(Stoeling,ANY-maze version 4.99 m)、震动切片机(Leica,VT1000S)、CMOS相机(HAMAMATSU,C11440-36U)、放大器(Axon Instrument, Multiclamp 700B)、数模转换器(Axon Instrument,1550B)、微电极拉制仪(Sutter, P-1000)。
